Objective:To investigate the effect of sildenafil citrate on the pharmacokinetics of sildenafil citrate and compare the results with those obtained with other drugs.
Design:The study was a randomized, double-blind, crossover study with a total of 12-week intervention. The study was carried out in 22 men aged 18 years and older with erectile dysfunction (ED) and/or premature ejaculation (PE). The study protocol was approved by the Ethics Committee of the Federal University of Sao Paulo, Brazil. The patients were randomly assigned to receive 100 mg sildenafil citrate or placebo at the beginning of the study. During the treatment period, patients were asked to report the mean erectile dysfunction and the mean premature ejaculation latency (PEL) at their first visit.
Participants:The study subjects were healthy volunteers. The study was conducted between September and October 2006. The volunteers completed the study in the form of a questionnaire and the subjects were instructed to answer the question “How do you feel after being sexually aroused for 12 hours?” During the follow-up period, the subjects completed the question “How is your erection and how long does it last?” at the beginning of the study and again at each visit. The patients were informed of the possible consequences of taking sildenafil citrate and were asked to report the time of ejaculation and the average time of sexual intercourse for the last two months.
Results:The results showed that sildenafil citrate significantly improved the erectile function and PEL in the patients. The mean erectile function was significantly improved by the study in patients who took sildenafil citrate and increased the mean PEL by more than 15%. The results for sildenafil citrate were also significantly improved in the patients who took sildenafil citrate, but the effect was not statistically significant. Sildenafil citrate also improved PEL in patients who did not take sildenafil citrate and did not significantly increase the mean PEL.
Conclusions:Our findings suggest that sildenafil citrate improves the pharmacokinetics of sildenafil citrate and that sildenafil citrate is effective in improving the ejaculatory control in patients with ED and PE.

Introduction
The phosphodiesterase 5 (PDE5) enzyme is involved in the regulation of erection. This enzyme is found within the cell membrane of various tissues, including the prostate, the testes and epididymis. Sildenafil citrate also improves the control of sildenafil citrate-induced erectile dysfunction and PE.

Materials and methods
